Polymerizing Actin Fibers Position Integrins Primed to Probe for Adhesion Sites

Migrating cells extend protrusions, probing the surroundingmatrix in search of permissive sites to form adhesions. We foundthat actin fibers polymerizing along the leading edge directedlocal protrusions and drove synchronous sideways movement ofß1 integrin adhesion receptors. These movements leadto the clustering and positioning of conformationally activated,but unligated, ß1 integrins along the leading edgeof fibroblast lamellae and growth cone filopodia. Thus, rapidactin-based movement of primed integrins along the leading edgesuggests a "sticky fingers" mechanism to probe for new adhesionsites and to direct migration.
