Members of the transforming growth factor (TGF) superfamily, such as Nodal, act as inducers for the formation of vertebrate mesoderm. In particular, Nodal is needed for normal embryo development, and its signaling must be tightly controlled. Zhang et al. (p. 114) report that Dapper2 modulates Nodal signaling activity in zebrafish embryos: loss of Dapper2 leads to up-regulation of Nodal-responsive genes in early gastrula, and conversely, overexpression of Dapper2 yields down-regulation of Nodal-responsive genes. Dapper2 coprecipitates with and disrupts the interactions of components of the TGF signaling pathway and promotes lysosomal TGF receptor degradation.
