Regulation of NF-B-Dependent Lymphocyte Activation and Development by Paracaspase

Paracaspase (MALT1), a member of an evolutionarily conservedsuperfamily of caspase-like proteins, has been shown to bindand colocalize with the protein Bcl10 in vitro and, becauseof this association, has been suggested to be involved in theCARMA1-Bcl10 pathway of antigen-induced nuclear factor B (NF-B)activation. We demonstrate that primary T and B lymphocytesfrom paracaspase-deficient mice are defective in antigen-receptorinducedNF-B activation, cytokine production, and proliferation. Paracaspaseacts downstream of Bcl10 to induce NF-B activation and is requiredfor the normal development of B cells, indicating that paracaspaseprovides the missing link between Bcl10 and activation of theIB kinase complex.
