NEUROSCIENCE:
An Accomplice for g-Secretase Brought into Focus

Sangram S. Sisodia

The presenilin proteins are mutated in some patients with early onset Alzheimer's disease, but the function of these proteins is not known. Accumulating evidence suggests that they are g-secretase enzymes that process the amyloid precursor protein. Now, as Sisodia discusses in this Perspective, another protein called nicastrin has been discovered that may contribute to the g-secretase activity of the presenilins.
