- Movie S1
Surface rendering of the S. typhimurium needle complex base (20mer). The absolute hand of the reconstruction was not determined. Time of animation: 18sec (for one full rotation).
- Movie S2
Conformational changes resulting from the assembly of the TTSS needle and inner rod.
The animation was created by morphing the contoured longitudinal sections shown in
Fig.3. No additional modeling was applied. Flexible regions that undergo conformational changes during reprogramming of the TTSS are highlighted in the superposition of still images (see the text for details). The most pronounced changes are observed in IR1 where the cup-like extension pushes down, and the socket-like feature moves outward. IR2 undergoes an inward clamping movement. Large conformational changes are also observed in OR1 and OR2, which are composed of the InvG protein. Overall, the changes result in a straightening of the outer lining of the internal cavity of the base and the creation of a new internal substructure, the inner rod, which connects the bottom of the base chamber to the insertion point of the needle at the top of the base.
