Before ovulation in mammals, a cascade of events resemblingan inflammatory and/or tissue remodeling process is triggeredby luteinizing hormone (LH) in the ovarian follicle. Many LHeffects, however, are thought to be indirect because of therestricted expression of its receptor. Here, we demonstratethat LH stimulation induces the transient and sequential expressionof the epidermal growth factor (EGF) family members amphiregulin,epiregulin, and beta-cellulin. Incubation of follicles withthese growth factors recapitulates the morphological and biochemicalevents triggered by LH, including cumulus expansion and oocytematuration. Thus, these EGF-related growth factors are paracrinemediators that propagate the LH signal throughout the follicle.
