Resurrecting the Ancestral Steroid Receptor: Ancient Origin of Estrogen Signaling

Receptors for sex and adrenal steroid hormones are absent fromfully sequenced invertebrate genomes and have not been recoveredfrom other invertebrates. Here we report the isolation of anestrogen receptor ortholog from the mollusk Aplysia californicaand the reconstruction, synthesis, and experimental characterizationof functional domains of the ancestral protein from which allextant steroid receptors (SRs) evolved. Our findings indicatethat SRs are extremely ancient and widespread, having diversifiedfrom a primordial gene before the origin of bilaterally symmetricanimals, and that this ancient receptor had estrogen receptorlikefunctionality. This gene was lost in the lineage leading toarthropods and nematodes and became independent of hormone regulationin the Aplysia lineage.
