Coordinate Regulation of Transcription and Splicing by Steroid Receptor Coregulators

Recent observations indicating that promoter identity influences
alternative RNA-processing decisions have created interestin the
regulatory interactions between RNA polymerase II transcriptionand
precursor messenger RNA (pre-mRNA) processing. We examinedthe
impact of steroid receptor-mediated transcription on RNA processingwith reporter genes subject to alternative splicing driven bysteroid-sensitive promoters. Steroid hormones affected the processingof pre-mRNA synthesized from steroid-sensitive promoters, butnot from
steroid-unresponsive promoters, in a steroid receptor-dependentand
receptor-selective manner. Several nuclear receptor coregulatorsshowed
differential splicing effects, suggesting that steroidhormone
receptors may simultaneously control gene transcriptionactivity and
exon content of the product mRNA by recruiting coregulatorsinvolved in
both processes.
