Enzymatic Synthesis of Tri- and Tetranucleotides of Defined Sequence

Conditions have been found under which polynucleotide phosphorylase adds only a few nucleotide residues to the 3' end of a dinucleotide primer. Pure tri- and tetranucleotides can be isolated from reaction mixtures in high yield.
